very hard to answer,

depends on what you mean, greatest by popularity, relevance, something else, etc....

here are a few answers:

gangsters paradise was fucking incredible when it first came out for both pop and rap, but i wouldn't consider it the greatest rap song ever.

honestly, in my opinion there are about 2-3 songs off enter the wu tang that could be in consideration.

i think we have to consider Pac, and my favorite of his is Every Thing They Owe Me (yeah yeah, white guilt whatever, its a bad ass song)

and while i don't think its the greatest ever, i'd put crossroads by bone thugs in the top 10

so many others...........there is no such thing as a greatest rap song ever. Some of the stuff Dre did, and Funk Flex put together needs to be considered. There are 100 I'm not thinking of. I didn't even get to half the groups. I kind of quit listening when Master P blew up and the whole evolution of hip hop diluted and then ruined the game. I didn't put any Jada, Luda, Em, Gangstarr, LL, Kwame, Craig Mack, Spice 1, The Geto Boys, Jay-Z, Mobb Deep, DJ Magic Mike, n2Deep, Kurtis Blow, The Phracyde, and countless others on there...

I've never listened to The Game, most of Jay-Z (not really a fan), dudes w/ Lil in their name, dudes w/ Yung in their name.... it's just old and been done before.
