so when ligands bind to two tyrosine-kinase receptor proteins, it activates their catalytic activity in the intracellular domain by dimerization (basically the hormone binds to the receptors causing the two receptors come together)

When the two receptors associate (touch) they activate eachother by phosphorylating specific tyrosine residues on their partner, a process referred to as autophosphorylation

The now activated tyrosine-kinase dimer binds docking proteins/exchange factors from the cytosol which stimulates the GDP (guanosine 5'-diphosphate) to be exchanged for GTP (guanosine 5'-triphosphate). The GTP then activates the tranducer Ras

The now active transducer Ras binds to and activates the effector enzyme Raf (MAPKKK)

The activation of Raf (MAPKKK) leads to a mitogen-activated protein kinase cascade [MAPKKK phosphorylates and activates MAPKK which then phosphorylates and activates MAPK]

activated MAPK, the secondary messenger, then enters the nuclear membrane and phosphorylates and activates certain transcription factors

Activated transcription factors bind to specific sites on the DNA and turn on/off the transcription of certain genes

Knopp's "Holes in the Bucket"
1) If no hormone is present, then the receptor proteins never dimerize, so they never activate the transducer which then can never activate the effector to activate the kinase cascade to make MAPK which is necessary in activating the transcription factors to turn genes on/off
2) GAP hydrolyzing GTP on Ras to GDP inactivates the transducer protein so that it is unable to activate the effector enzyme
3) Kinase cascade and transcription factors can become deactived by phosphatases which de-phosphorylate them
